چکیده

جاذبه از مهمترین نیروهای وارده بر موجودات زنده روی زمین است که در طول حیات، بر سلول­ها، مولکول­ها و در نتیجه بر کل ارگانیسم تاثیرگذار است. هدف از این مطالعه، بررسی اثر بی وزنی شبیه سازی شده بر تغییرات بیان ژن سرکوب گر متاستاز RKIp در رده سلولی MDA-MB-231 سرطان سینه می­باشد.
مواد و روش ­ها: به منظور اعمال بی وزنی، سلول­های سرطانی، به مدت یک روز و سه روز بر روی دستگاه کلینواستت قرار گرفتند. سپس RNA از سلول­ ها استخراج و تغییرات بیان ژن RKIp توسط qReal time PCR سنجش گردید.
نتایج: یافته ­ها نشان دادند که بی­وزنی به مدت یک روز، سطح بیان ژن RKIp را کاهش داده اما با ادامه اعمال بی وزنی تا سه روز، بیان به سطح کنترل بازگشته است.
نتیجه گیری: با بهینه سازی مدت زمان تیمار بی وزنی احتمالاً شاهد تغییرات قابل توجه در بیان ژن RKIp بوده تا از این طریق بتوان گام­هایی را در کشف مکانیسم ­های درگیر در سرطان و درمان آن برداشت.

Introduction: Gravity is one of the most important forces exposed to the organisms on the earth which affects the cells, molecules and thus the entire of organism. The purpose of this study is to investigate the effect of the simulated microgravity on the changes of RKIP metastasis suppressor gene expression on MDA-MB-231 breast cancer cell line.
Materials and Methods: In order to impose microgravity state, cancer cells were placed on clinostat for one and three days. Then RNA was extracted from the cells and RKIP gene expression changes were evaluated by qReal time PCR.
Results: The findings showed that the microgravity during one day reduced the RKIP expression level, but with continuing up to three days, the expression returned to the control level.
Conclusion: By optimizing the duration of microgravity, it can be likely observed the significant effects on the RKIP gene expression changes and so that useful steps can be taken to discover the cancer mechanisms and its treatment.
